Indian biotechnology company Immunoscript Life Science has secured up to US$8.9 million in funding from the Coalition for Epidemic Preparedness Innovations (CEPI) to advance clinical testing of a next-generation RNA vaccine platform that could eliminate one of the biggest challenges facing modern vaccination programmes: the cold chain.<\/p>\n<p class=\"wp-block-paragraph\">The investment is significant not only for India\u2019s rapidly expanding biotechnology sector but also for global public health. If successful, the technology could help deliver RNA vaccines to remote and low-resource communities that currently lack the ultra-low temperature storage infrastructure required by many conventional mRNA vaccines. At a time when public health authorities are increasingly concerned about emerging infectious diseases, so-called \u201cDisease X\u201d threats and the possibility of future pandemics, advances that improve vaccine accessibility may prove just as important as advances in vaccine efficacy.<\/p>\n<p class=\"wp-block-paragraph\">Messenger RNA (mRNA) vaccines became one of the defining scientific successes of the COVID-19 pandemic. Companies such as Pfizer-BioNTech and Moderna demonstrated that RNA-based vaccines could be designed, manufactured and deployed far more rapidly than many traditional vaccine technologies. The advantages of RNA vaccines are well established. Unlike conventional vaccine approaches that may require growing pathogens or producing large quantities of proteins, RNA platforms can be adapted relatively quickly once the genetic sequence of a new pathogen becomes available.<\/p>\n<p class=\"wp-block-paragraph\">This speed forms the foundation of CEPI\u2019s ambitious 100 Days Mission, a global initiative supported by G7 and G20 nations, including India, that aims to develop safe and effective vaccines against future epidemic threats within 100 days of identifying a new pathogen.<\/p>\n<p class=\"wp-block-paragraph\">Yet the COVID-19 response also highlighted a major weakness of existing RNA vaccine technologies. Many formulations require storage and transportation at extremely low temperatures. Maintaining such cold-chain systems is expensive, technically challenging and often impractical in rural regions and lower-income countries. <em>The result is a paradox.<\/em> Some of the world\u2019s most advanced vaccine technologies remain difficult to deploy in the places where they may be needed most.<\/p>\n<h2 id=\"h-a-different-approach-to-rna-vaccines\" class=\"wp-block-heading\"><strong>A different approach to RNA vaccines<\/strong><\/h2>\n<p class=\"wp-block-paragraph\">Immunoscript\u2019s proprietary CLNE\u2122 self-amplifying RNA (saRNA) platform seeks to overcome this obstacle. Traditional mRNA vaccines typically encapsulate RNA molecules within lipid nanoparticles. The CLNE platform instead binds RNA directly to the outer surface of nanoemulsion droplets, creating a system that can be dried into a powder and stored under conventional refrigeration conditions of 2\u00b0C to 8\u00b0C.<\/p>\n<p class=\"wp-block-paragraph\">This distinction is important for outbreak preparedness. A vaccine that can be transported through routine healthcare supply chains rather than specialised cryogenic systems could substantially improve global access. In an emergency, millions of doses could potentially be distributed more rapidly across regions where cold-chain infrastructure remains limited.<\/p>\n<p class=\"wp-block-paragraph\">According to CEPI Chief Executive Officer Dr. Richard Hatchett, equitable vaccine access remains a priority for global preparedness efforts. The organisation views thermostable RNA vaccines as a potentially important tool in overcoming one of the major barriers to broader deployment of RNA vaccine technologies during outbreaks.<\/p>\n<p class=\"wp-block-paragraph\">The importance of such technologies extends well beyond COVID-19. The global health community continues to monitor a wide range of emerging infectious disease threats. Recent years have seen outbreaks involving Mpox, Ebola virus disease, Nipah virus and other pathogens with epidemic potential. CEPI\u2019s portfolio includes programmes addressing multiple emerging diseases while simultaneously investing in platform technologies designed to accelerate future vaccine development.<\/p>\n<p class=\"wp-block-paragraph\">The challenge facing public health agencies is that it is impossible to predict with certainty which pathogen will trigger the next major outbreak. This is where platform technologies become increasingly valuable. Instead of building a new vaccine production process from scratch for every threat, advanced RNA systems may provide a flexible foundation that can be adapted rapidly once a pathogen has been identified and sequenced. The goal is not merely faster response but a fundamentally different model of pandemic preparedness.<\/p>\n<h2 id=\"h-why-self-amplifying-rna-matters\" class=\"wp-block-heading\"><strong>Why self-amplifying RNA matters<\/strong><\/h2>\n<p class=\"wp-block-paragraph\">The CLNE platform incorporates another notable feature: self-amplifying RNA. Unlike conventional mRNA vaccines, self-amplifying RNA technologies effectively instruct cells to generate additional copies of the vaccine RNA after administration. In practical terms, this means smaller quantities of starting material may produce a robust immune response.<\/p>\n<p class=\"wp-block-paragraph\">From a public health perspective, this offers several advantages. First, manufacturing capacity can potentially be extended during emergencies, enabling larger numbers of vaccine doses to be produced from the same manufacturing infrastructure. Second, lower production requirements may reduce overall costs. Third, the technology could improve preparedness for large-scale outbreak scenarios where demand surges rapidly. Researchers have increasingly identified self-amplifying RNA as one of the most promising next-generation vaccine technologies currently under development.<\/p>\n<p class=\"wp-block-paragraph\">The new CEPI funding will support an early-stage clinical study evaluating both the safety of the CLNE platform and the immune responses generated by a vaccine candidate targeting rabies. Rabies serves as a useful prototype disease because it remains a significant public health concern in many parts of the world while also providing an opportunity to demonstrate the platform\u2019s potential beyond COVID-19 applications.<\/p>\n<p class=\"wp-block-paragraph\">Researchers plan to use artificial intelligence tools to optimise dose selection during the study, illustrating how digital technologies are increasingly being integrated into vaccine development programmes. This combination of RNA science, computational biology and advanced manufacturing reflects broader trends across modern biotechnology.<\/p>\n<h2 id=\"h-india-s-growing-biotechnology-influence\" class=\"wp-block-heading\"><strong>India\u2019s growing biotechnology influence<\/strong><\/h2>\n<p class=\"wp-block-paragraph\">The development also highlights India\u2019s expanding role within the global biotechnology and vaccine ecosystem. India is already one of the world\u2019s most important vaccine manufacturing centres, supplying large quantities of vaccines to domestic and international markets. Increasing investment in advanced platform technologies may further strengthen the country\u2019s position as a global innovation hub.<\/p>\n<p class=\"wp-block-paragraph\">Immunoscript itself represents part of a broader evolution of the Indian life sciences sector towards cutting-edge genomic medicine, advanced therapeutics and precision healthcare technologies. The company describes its mission as using biological engineering, artificial intelligence and translational science to address global healthcare challenges. For India, success in next-generation RNA technologies could deliver both economic and strategic benefits while contributing to international health security.<\/p>\n<p class=\"wp-block-paragraph\">Another aspect of the project is that its potential extends beyond future pandemics. Although much attention is focused on Disease X scenarios, thermostable RNA platforms may also support vaccine development against existing diseases that continue to impose major burdens on public health systems around the world. In theory, technologies that simplify manufacturing, distribution and storage could expand access not only during emergencies but also during routine immunisation programmes.
